Need Robust HPLC Methods ?
Trust OSIRIS, the Interactive Assistant
OSIRIS is a software tool dedicated to method development and optimisation in Liquid Chromatography. Due to its unsurpassed algorithms based on 10 years of research in the Analytical Sciences Laboratory in Lyon University, OSIRIS allows you to set up robust methods in a very short time and after only a few runs.
OSIRIS is a full 32 bit application running under Windows 98, NT, 2000 and XP. It has been developed using the latest software technology and following industry standards. But it also offers a very friendly to use interface including a unique Assistant which guides the analyst step-by-step during the Optimization process.
OSIRIS is the ideal tool to:
- Determine good separation conditions quickly and accurately
- Improve the ruggedness of an HPLC method
- Reduce the analysis time of an existing method,
- Teach HPLC and the effects of the different parameters.
Many HPLC method development laboratories, in particular in pharmaceutical industry, already use OSIRIS and have experienced its reliability. Evaluation version (trial mode) is a fully functional program that is active for 30-days (only save and print are not possible). The demonstration guide (Quick Tour) contains basics information and describes few available examples.
Develop
OSIRIS speeds up the development of your HPLC separations in isocratic mode as well as for gradient operation. Usually, the set-up of a complete method requires time-consuming analysis in your laboratory before obtaining a satisfactory result. With a minimum of experimentation OSIRIS is able to model retention behaviour of compounds and then to offer computer predictions of various chromatograms for different analytical conditions. Using this predictive modelling, a wide range of conditions can be simulated in only few minutes.
The Wizard guides you during the development of the method :

The chromatographic behavior of the solutes is modelled as a function of the elution conditions. This model is obtained from the results and the chromatographic data of a few preliminary runs. OSIRIS guides the user by proposing conditions for these preliminary runs. The number of runs and the conditions depends on the type of optimization process :
- Mobile phase composition (isocratic, binary, ternary or quaternary, linear and multi-step gradient)
- pH
- Temperature
- Mobile phase/pH.
- Mobile phase/Temperature
Optimize
OSIRIS can also improve your existing methods.
You can fix your own criteria in terms of quality of separation (resolution), analysis time and/or robustness. You can evaluate how sensitive this method is to small variations in the conditions or to shorten the analysis time without sacrificing resolution. OSIRIS allows you also to focus on the separation of target compounds.

to select the most favorable composition zones
for ternary and quaternary mobile phases. |
Experiment
OSIRIS accurately models the effect of different parameters on your separation :
- Organic solvent concentration (binary system)
- Second organic solvent concentration (ternary system)
- Third organic solvent concentration (isocratic quaternary system)
- Temperature
- pH
- Gradient conditions and profile
This modelling can be performed simultaneously for up to 2 or 3 parameters. The mathematical models used are specific for each type of parameters and enables the prediction of an exact optimum.

The Gradient Editor allows to define some gradient profiles and to predict the resulting chromatogram.
You can change the initial and the final points of the gradient just by dragging and dropping them with
the mouse or by entering values for initial %B and gradient slope in the table :

Validate
OSIRIS is a very helpful tool for method transfer and validation since it takes into account the robustness of the analytical conditions and the dwell volume of your chromatographic equipment. The separation could be modelized versus the resolution but also integrating the Robustness in the Response function.

You can also study the effect of modifications in the physical properties of the column. Thanks to its Simulation table, OSIRIS allows you to store and to compare chromatograms obtained using different conditions. Finally, OSIRIS enables you to fully document your method and to create a detailed report.
